visit to the Deplanck Laboratory EPFL
At the Deplancke lab, we leverage nano- and microtechnology for multimodal single-cell analysis, integrating imaging with transcriptomics and time-resolved transcriptomics. To seamlessly couple imaging with transcriptomics, we have developed a custom instrument that combines microscopy with deterministic cell handling, as presented in Bues, Biocanin et al. (2022). This system enables precise pairing of imaging and transcriptomic data at the single-cell level.
For temporal profiling of live cells, we employ atomic force microscopy (AFM), specifically FluidFM, to extract cytoplasmic material in a minimally invasive manner. This technique, known as Live-seq, allows for time-resolved transcriptomic analysis without compromising cell viability, as demonstrated in Chen, Orane Guillaume-Gentil et al. (2022). By preserving cellular integrity while capturing dynamic gene expression changes, Live-seq provides unique insights into cellular states and heterogeneity.
